Ensure antibody titer and cross-reactivity

1. Antigen Validation and Enzymatic Digestion (1 week)

- SDS-PAGE for drug purity assessment; UV spectroscopy for drug concentration measurement.

- Enzymatic digestion of intact antibody drugs into F(ab')₂ fragments to analyze purity and activity.

2. Immunization and Serum Titer Determination (6 weeks)

- Conduct 3-4 immunizations within 35–42 days.Achieve serum titer >1:1,000,000.

3. Antibody Purification (1 week)

- Antigen Affinity Chromatography； Total Human IgG purification.

4. Anti-Drug Antibody (ADA) Positive Detection (1 week)

- Purity/Concentration: SDS-PAGE for drug purity; UV spectroscopy for drug concentration.

- ELISA Analysis: Confirm dilution >1:500,000. Ensure <2% cross-reactivity.
